ABSTRACT

Objective To investigate the expression and clinical pathological correlation of STAT3 and E-cadherin in esophageal squamous cell carcinoma,as well as the effect of the process in the transformation of epithelial mesenchymal transition of esophageal squamous cell carcinoma.Methods The 50 cases of esophageal squamous cell carcinoma tissues and corresponding adjacent tissues,to detect the expression of STAT3 and E-cadherin expression by RT-qPCR method in carcinoma and adjacent normal tissue expression that differential expression in mRNA level,analyzed its relationship with clinical pathological characteristics between patients;the expression of STAT3 and E-cadherin was detected by Western blotting in 4 cases of esophageal squamous cell carcinoma by simple random sampling.The author detected the expression of STAT3 and E-cadherin in three strains of esophageal squamous cell carcinoma cell lines using RT-qPCR and Western blotting,in which a high expression strains were transfected by STAT3 small interfering RNA screening,as the experimental group,using double stranded nonsense RNA transfection as the control group,detected the changes of expression level of STAT3 and E-cadherin.Detected cell migration and invasion by cell scratch test and Transwell cell invasion test in the experimental group.Measurement data were expressed by ((x) ± s),and the t test was used comparison between groups.Results In 50 cases of esophageal carcinoma,the relative expression of STAT3 was 1.87 ± 0.71,E-cadherin was 0.63 ± 0.28,in paracancerous tissue STAT3 was 0.56 ± 0.45,E-cadherin was 2.01 ± 0.68;analysis of clinical pathological features of esophageal squamous cell carcinoma showed relatively high expression of E-cadherin as relatively low expression levels of STAT3,related to the depth and lymph node metastasis in carcinoma tissues (P < 0.05).Among which 4 cases were selected,the expression of STAT3 in cancer tissues higher than in corresponding adjacent normal tissues,0.28 ± 0.06 vs 0.05 ± 0.02,P < 0.05,and the expression of E-cadherin in cancer tissues was significantly lower than that in adjacent normal tissues,0.27 ± 0.07,0.59 ± 0.09,P < 0.05.The high expression of EC109 from carcinoma cell lines was as experimentation of esophageal squamous cell,RT-qPCR results showed that the expression of STAT3 was significantly decreased in small interfering RNA transfection group than the control group 0.85 ± 0.24 vs 3.01 ± 0.20,P < 0.05,while up-regulation of E-cadherin protein expression,2.69 ± 0.29 vs 0.36 ± 0.16,P < 0.05;Western blotting detection,compared with the control group,the expression of STAT3 protein was successfully inhibited after transfection of STAT3 small interference RNA fragment (0.09 ±0.04 vs 0.32 ±0.18,P < 0.05),while the expression of E-cadherin was up-regulated in EC109 cells (0.75 ±0.11 vs 0.15 ±0.05,P <0.05).After downregulation of STAT3 expression,cell scratch test and transwell cell invasion test showed that the migration and the number ofinvasion cell number of STAT3 small interfering RNA transfection group in esophageal squamous carcinoma cell line EC109 compared with the control transfected double-stranded non-sense RNA group were decreased,so the downregulation of STAT3 expression after migration,invasion ability were weakened (P < 0.05).Conclusion In esophageal squamous cell carcinoma,STAT3 transcription factor can mediate E-cadherin expression and affect epithelial mesenchymal transition phenotype,thereby affecting tumor migration and invasion.

ABSTRACT

Objective To investigate the effect of acupuncture combined with human amniotic mesenchymal stem cells ( HAMCs ) transplantation on the osteoporosis in ovariectomized rats.Methods Forty-five healthy female non-pregnant Wistar rats were randomly divided into sham operated group, model group and experimental group ( acupuncture+HAMC transplantation), 15 rats in each group.At 7 days after the surgical wound healing, the rats received acupuncture every day and intravenous injection of HAMCs suspension ( except the sham operated group) once a week for consecutive 12 weeks.The body weight was measured every week.24 hours after the last intervention, the rats were killed and specimens were collected.Specimens of the mid-, proximal-and distal femur were taken to measure the bone mineral density using a dual energy X-ray absorptiometer.At 12 weeks after the intervention, changes of the expression of serum 25-hydroxy-vitamin D (25-HVD), C-terminal peptide of type I collagen (CTX-I), bone specific alkaline phosphatase ( BAP) and human tartrate-resistant acid phosphatase 5b ( TRACP 5b) were determined by ELISA, the biomechanical properties of the removed femur was measured, and the expression of transforming growth factor-β( TGF-β) in the vertebrae was assessed by RT-PCR assay.Results Body weight of rats in the sham operation group was increased gradually in accordance with the natural growth of animals, that of the model group was significantly higher than that of the sham operated group since two months after modeling, and the body weight of the experimental group was similar to that of the sham operated group.The bone mineral density and calcium content of the model group were significantly decreased compared with that of the sham operated group ( P<0.05) , and the bone mineral density and bone calcium content of the experimental group were significantly higher than that of the model group ( P <0.05 ) .The ELISA showed that the expressions of serum 25-HVD, CTX I, BAP, and TRAP5b in the model group were significantly lower than that of the sham operated group, and those of the experimental group were significantly higher than that of the model group ( P<0.05 ) . Measurement of biomechanical properties showed that the limit load, limit stress and elastic modulus of the femur of the sham operated group were significantly higher than that of the model group ( P<0.05 ) , and the limit load and elastic modulus of the experimental group were significantly higher than that of the sham operated group ( P <0.05 ) .T-PCR showed that the expression of TGF-β1 m-RNA in the experimental group was significantly up-regulated than that of the sham operated and model groups ( P<0.05) .Conclusions Acupuncture combined with human amniotic mesenchymal stem cell ( HAMCs) transplantation has a synergistic effect on the treatment of osteoporosis, and can improve the conditions in ovariectomized rats.

ABSTRACT

Objective To investigate the short-term clinical efficacy and complication of esophageal cancer patients with two-field lymph node dissection by thoracolaparoscopic esophagectomy surgery and open surgery. Methods One hundred and fifty esophageal cancer patients with two-field lymph node dissection were selected, and they were divided into control group (using open surgery, 75 cases) and observation group (using thoracolaparoscopic esophagectomy surgery, 75 cases) by random digits table method. The operation time, bleeding amount, hospital staying time, number of lymph node dissection, reoperation rate, intensive care unit (ICU) transferring rate and postoperative complication were compared. Results There was no statistical difference in operation time between 2 groups ( P>0.05). The bleeding amount and hospital staying time in observation group were significantly lower than those in control group: (210.33 ± 30.71) ml vs. (254.59±35.28) ml and (8.45±1.52) d vs. (11.61±2.08) d, there were statistical differences (P0.05). There were no statistical differences in incidences of hoarseness and anastomotic stenosis between 2 groups ( P>0.05). The incidences of pulmonary infection and arrhythmia in observation group were significantly lower than those in control group:17.33%(13/75) vs. 30.67%(23/75) and 2.67%(2/75) vs. 14.67%(11/75), and there were statistical differences (P<0.05). Conclusion Compared with open surgery, thoracolaparoscopic esophagectomy surgery with two-field lymph node dissection for esophageal cancer patients can effectively reduce the degree of operative trauma, accelerate postoperative rehabilitation process, improve the effects of lymph node dissection, and reduce postoperative complication risk.

ABSTRACT

Objective To evaluate the clinical efficacy of different treatments on non-small cell lung cancer (NSCLC) patients with brain metastases and to explore the influential factors of the prognosis.Methods The NSCLC patients with brain metastases treated from Jan.2010 to Dec.2011 were follow-up.The survival time and influences resulted from the treatments were analyzed.Results The average survive time of these patients was (11.93±5.53) months,and the median survive time was 11 months.The 6-month,1-year and 2-year overall survival rates were 90.7 %,41.1% and 6.4 %,respectively.Multivariate analysis showed that control of extracranial lesions,Kamofsky score,target therapy and age were independent predictive factors of survival,and the OR value were 0.358 (95 % CI0.217-0.593),0.302 (95 % CI 0.182-0.502),0.170 (95 % CI 0.098-0.296) and 1.635 (95 % CI 1.010-2.647),respectively (all P < 0.01).Conclusions Radiation therapy is an effective treatment on non-small cell lung cancer with brain metastases.Biological target therapy can effectively improve survival.The survival time also is correlated with age,Karnofsky score and control of extracranial lesions.

ABSTRACT

Objective To establ ish the chromatogrphic fingerprint of Jisheng Shenqi Pills by HPLC method for evaluating the quality of the preparation. Methods The mobile phase was acetonitrile-0.3%acetic acid solution in a gradient elution mode.The flow rate was 1.0 ml/min and the UV detection wavelength was 240 nm. The similarity of 7 batches of samples was evaluated by using Similarity Calculation Software of TCM Chromatogrphic Fingerprint (2004A edition). Results Nine common peaks were identified in 7 batches of samples and found belonging to their single medicinal material. The similarity of fingerprint was all higher than 0.797. Conclusion The method is simple and accurate,reproducible for quality control of the preparation.

ABSTRACT

Objective To establish an HPLC method for determination of low-molecular-weight carbonyl compounds in the oil-containing herbs. Methods After carbonyl compounds in the samples were extracted with water, the solution reacted with 2, 4-dinitrophenylhydrazine (DNPH) in an acidic medium to form 2, 4-dinitrophenylhydrazone derivatives, which were separated on Kromasil KR100-5 C18 column (250 mm× 4.6 mm, 5 μm). The mobile phase A was water-acetonitrile-tetrahydrofuran-isopropanol (59∶30∶10∶1), mobile phase B was water-acetonitrile (35∶65), gradient elution. The flow rate was 0.8 mL/min, detection wavelength was 365 nm, and column temperature was 30 ℃. Results Good linearities were obtained in corresponding concentration ranges, with correlation coefficient over 0.999. The limits of detection of the eight DNPH derivatives were 0.002-0.008 μg/mL, and the average recoveries were 88.49%-93.65%. Conclusion The method is simple, accurate and reliable, with good reproducibility.

Based on its years of experiences,Shenzhen began to openly enroll medical school graduates nationwide since 2010 for standardized residents training.This started the brand new training of resident physicians,which enjoys unreserved policy and budget support from the government.Thanks to training support from a number of medical schools and teaching hospitals with prime resources in the country,training quality is maintained at a high level.As one of the first cities in practicing the standardized residents training,Shenzhen has made an initial success.But the management system and standardized training model still need further improvement and further exploration to improve the training program in the city.

ABSTRACT

Objective To investigate the morbility and epidemic characteristics of hair dye dermatitis in individuals who dyed their hair in Tianjin.Methods Questionnaires were distributed to the outpatients in the Gerneral Hospital of Tianjin Medical University,students and teachers in Tianjin Medical University,residents in the community and customers in barber shop from Aug.2007 to Mar.2008.The personal data including the methods and site of coloring hair and something correlated to hair dyes were investigated.Results A total of 597 cases with the history of coloring hair were enrolled in the study,including 485 women and 112 men,with mean age of 41 years (ranged from 16- 74years).Among 597 cases,69 cases had allergic reactions to hair dye,including 51 women and 18 men,with mean age of 44 years (ranged from 19-65 years).The median age of the first coloring hair was 40 years (ranged from 3-50 years).The comparison between the sites of coloring hair had no statistic significance (P >0.05),but there was a significant difference between black dyed hairs and col or dyed hairs (P <0.05).Conclusions People with black hair dye are prone to be allergic.

ABSTRACT

Objective:To investigate the effect of ethyl acetate extract from Cortex periplocae (CPEAE) on apoptosis of human esophageal carcinoma cell line TE-13 and to elucidate its mechanism. Methods:Inhibitory effect of CPEAE on TE-13 proliferation was tested by MTT assay. The morphological changes of cell apoptosis were observed by Giemsa staining and transmission electron microscopy. Cell cycle and apoptotic ratio were tested by flow cytometry (FCM). The protein expression of CDK4 was observed by Western blotting.Results:CPEAE inhibited proliferation of TE-13 cells in a concentration-dependent and time-dependent manner, and its IC_(50) value was (2.443±0.005) μg/mL at 48 h (P<0.05). The characteristic morphological changes of apoptosis were observed in TE-13 cells after treatment with CPEAE under transmission microscope. A typical subdiploid peak was detected by flow cytometry. CPEAE decreased the expression of gene CDK4 in TE-13 cells. Conclusion:CPEAE can induce apoptosis of TE-13 cells. The effect is related with down-regulation of CDK4 expression.

ABSTRACT

This study was designed to detect the effects of Bazhen decoction on the proliferation and activation of T lymphocytes. T lymphocytes were isolated. The effects of Bazhen decoction on the prolifertion and activation of T lymphocytes, and on the secretion of IFN-gamma IL-2 from T lymphocytes, were detected by MTT, Flow cytometry and ELISA. Results showed that proliferation of T lymphocytes was promoted significantly by different concentration of Bazhen decoction; and after different time, the relationships of "the longer the time, the higher the concentration, and the more enhanced the proliferation" came to be apparent. After 72h, T lymphocytes were activated with different concentration of Bazhen decoction, the rate of CD69+ T cells increased signficantly, the secretion of IFN-gamma and IL-2 increased signficicantly, and the effect exhibited a dose-dependent manner. The results of this epxeriment indicated that Bazhen decoction could promote the proliferation and activation of T lymphocytes.

ABSTRACT

Objective To establish a capillary gas chromatography method for the determination of menthol and methyl salicylate in Shangtongning adhesive plaster. Methods The internal standard substance of n-Pentadecane was employed. The GC system consisted of a DB-WAX capillary column (30 m?0.32 mm, 0.25 ?m), nitrogen as the carrier gas, column temperature at 130 ℃, FID as the detector at 200 ℃, and with a split ratio of 12∶1. Results The contents of menthol and methyl salicylate in 6 lots of samples were determined and the constituents concerned were separated well under the chromatographic condition. The average recovery of menthol and methyl salicylate were 101.52% (RSD=2.66%) and 100.34% (RSD=3.38%), respectively. Conclusion The method was sensitive, accurate, specific and suitable for the quality control of the preparation.
